| D-泛解酸内酯(D-pantolactone,D-PL)是一种重要的手性中间体,主要用于合成D-泛酸即维生素B5及其衍生物。目前,全球对于D-泛解酸内酯的需求量仍在不断上升,因此开发高效、环保的生产路线具有重要意义。酶法合成D-泛解酸内酯被认为是一条具有广阔生产前景的合成路线,具有绿色、环保等特点,也是目前工业生产的主要方法。本文综述近年来酶法合成D-泛解酸内酯的研究进展,讨论了不同反应条件下D-泛解酸内酯水解酶(D-lactono hydrolase,D-Lac)的酶学性质、表达与定向进化,以及不对称还原酮基泛解酸内酯的研究进展,比较了化学法、直接发酵法与生物酶法的优势,为后续设计生产D-泛解酸内酯及其衍生物的方法提供参考。 |

| D-Pantolactone serves as a pivotal chiral intermediate in the synthesis of D-pantothenic acid (vitamin B5) and its derivative compounds. Given the escalating global demand for D-pantolactone, the development of efficient and environmentally benign production routes has emerged as a critical research focus. Enzymatic synthesis of D-pantolactone stands out as a highly promising approach, distinguished by its green and eco-friendly attributes, and has become the predominant method in industrial production. This comprehensive review delves into the recent advancements in the enzymatic synthesis of D-pantolactone. It meticulously examines the enzymatic properties, expression, and directed evolution of D-pantolactone hydrolase under different experimental conditions. Additionally, it explores the latest progress in the asymmetric reduction of keto-pantolactone. By comparing the merits of various synthesis methods, this review offers valuable insights and guidance for the future design of production strategies for D-pantolactone and its derivatives. |
