      日益严峻的抗生素耐药性现象已经成为危害世界公共卫生安全的巨大隐患,寻找结构新颖、活性良好的抗生素是全球所面临的共同挑战。近年来,越来越多的含邻烷基肉桂酰结构单元天然产物被发现,并因其独特的结构以及广泛而显著的生物活性而受到关注。这些化合物大部分具有抗菌、抗肿瘤和抗结核等多种生物活性,是重要的药物先导物,具有广阔的研究前景。本文首先在结构和活性上对2024年及之前报道的该类化合物进行归类总结;然后就邻烷基肉桂酰结构单元相关生物合成分子机制研究进展进行了综述,包括碳骨架组装过程、苯环形成以及二聚化反应机制;最后对含邻烷基肉桂酰结构单元活性天然产物的靶向发现策略进行了描述,并对未来的发展方向进行了展望,以期为相关系统性研究工作的开展提供参考。
英文摘要:
      The increasingly antibiotic resistance has become a significant threat to global public health security. The discovery of antibiotics with novel structures and good activity is a shared challenge confronting the global community. In recent years, a growing number of ortho -alkyl cinnamoyl lipids ( o -CL) moiety containing natural products have been identified, which have attracted more and more attention due to their unique structures and significant biological activities. Most of these compounds exhibit diverse bioactivities, such as antibacterial, antitumor, and antitubercular properties, making them important drug leads with extensive research potential. This article summarizes the structures and bioactivities of these compounds reported up to 2024. It also provides an overview of the advances in understanding the molecular mechanisms underlying the biosynthesis of o -CL, including the carbon skeleton assembly, aromatic ring formation, and dimerization processes. Furthermore, the article discusses targeted discovery strategies for bioactive natural products containing o -CL moiety. An outlook on future development directions is also indicated. This review would serve as a valuable reference for the research field of nature product discovery.
