      针对在使用试管扩散法检测食品中抗生素残留总量时出现的检测管制备时间长和保质期短的问题,介绍了一种简单高效的芽胞纯化方法。以嗜热脂肪地芽胞杆菌(Geobacillus stearothermophilus CICC 10392)为例,应用此方法对其芽胞进行纯化后,通过镜检等方式从纯化后芽胞悬液的纯度和保藏2 a后芽胞悬液的质量等方面对此方法进行了评价。结果显示,纯化后芽胞悬液中几乎全部为游离的芽胞,只有个别已萌发的芽胞混在其中;保藏2 a后,芽胞悬液中的大部分芽胞仍为游离的芽胞,只有少量芽胞萌发;芽胞悬液在600 nm处的吸光度值与其菌落数的相关系数为0.99;通过此方法制备的其他三种芽胞杆菌的芽胞悬液中几乎全部为游离的芽胞,只有个别细胞碎屑混在其中;与其他方法相比,此方法操作较简单,所需仪器较少,不使用任何试剂且耗时更短。

      A simple and efficient gemmae purification method was introduced in this study aiming at the problem existed when using test tube diffusion method to detect the amount of antibiotic residues in food products, due to long preparation time and short shelf life of the testing tubes. Geobacillus stearothermophilus CICC 10392 was taken as an example to demonstrate this method. The effects of this method were evaluated by examining the purity of the gemmae suspension after purification and the quality of the gemmae suspension after two years′ storage, through microscopy and other measures. The results showed that after purification, almost all the gemmae in the suspension were free gemmae and only a few germinated gemmae were mixed in the suspension. After two years′ storage, most of the gemmae in the suspension were still free gemmae and only a few gemmae had germinated. The correlation coefficient between absorbance value at 600 nm and their colony amount of the gemmae suspension was at 099. In the gemmae suspensions of three other gemmae producing strains, almost all gemmae were free gemmae and only a few cell debris were mixed in the suspension. The process of this method was relatively simple, and only some instruments were required, no reagent was needed and the processing time was shorter than the other purification methods.
