      淀粉样β蛋白质(Amyloid-β protein, Aβ)的错误折叠和聚集形成多种形式的毒性各异的聚集体是阿尔茨海默病(Alzheimer′s disease, AD)发生和发展的主要原因之一。研究Aβ的错误折叠和聚集机制及开发高效的抑制剂对AD的治疗至关重要。上述研究均需要大量的高纯度Aβ,而基于基因工程技术的重组蛋白质生产是获得Aβ及其突变体的常用工具。基于近年来Aβ微生物表达相关领域取得的进展,结合作者的相关研究,介绍了Aβ的微生物表达和纯化以及体内聚集抑制剂筛选系统的构建及使用,并讨论其优点和局限性,为淀粉样蛋白沉积疾病的分子机理和抑制剂开发的相关研究提供理论依据和技术支撑。
英文摘要:
      Misfolding and fibrillogenesis of Amyloid-β proteins (Aβ) into different kinds of aggregates associated with cytotoxicity is one of the main reasons of the occurrence and development of Alzheimer′s disease (AD). The study of the misfolding and aggregation mechanism of Aβ and the development of effective inhibitors against fibrillogenesis are essential for the treatment of AD, which are needed a large amounts of high purity of Aβ. Genetic engineering technology was used to produce large amounts of recombinant Aβ and its mutants. Based on recent advances in the field of Aβ microbial expression and combined with the authors′ relevant studies, biological expression of Aβ, the construction of the in vivo screening systems and its application were introduced in this review. And their advantages and disadvantages were also discussed. It would provide theoretical basis and technical support for the research on the molecular mechanism of the aggregation of amyloid proteins and the development of novel inhibitors.
