      探讨circRALB对宫颈癌细胞株Hela细胞生物学行为的影响。化学合成circRALB干扰序列si_RALB以及无关序列si_nc,瞬时转染Hela细胞。荧光定量PCR法检测干扰序列si_RALB对Hela细胞中circRALB表达的影响;细胞计数、MTS法、流式细胞术、细胞划痕实验和Transwell实验检测circRALB对Hela细胞的增殖、 凋亡、 迁移和侵袭能力的影响。结果显示,瞬时转染效率为(89.67±3.1)%。干扰序列si_RALB转染Hela细胞,circRALB表达量为si_nc组的(47.3±1.5)%,差异有统计学意义;si_RALB转染组细胞增殖活性明显低于si_nc对照组,差异有统计学意义;si_RALB转染组细胞凋亡率为(9.17±1.17)%,si_nc对照组细胞凋亡率为(7.2±0.32)%,空白对照组细胞凋亡率(5.63±0.31)%,尽管si_RALB细胞凋亡率大于si_nc对照组细胞凋亡率,但差异无统计学意义;si_RALB转染组与si_nc转染组相比,划痕间距缩小不明显;si_RALB转染组Hela细胞穿膜数为(21.33±2.19)个,显著低于si_nc组(45.33±1.76)个和空白对照组(52±153)个。化学合成的circRALB干扰序列转染Hela细胞后,能显著降低细胞中circRALB的表达量。CircRALB表达下调可显著抑制宫颈癌细胞系Hela细胞的增殖、迁移和侵袭能力,但对其凋亡无明显影响。结果提示CircRALB的表达与宫颈癌细胞的生物学行为有关,可能成为有效的宫颈癌治疗靶标。

      The effect of circRALB in cervical cancer cell line Hela cells on the biological behavior of tumor cells was investigated. CircRALB interfering sequence as well as si_nc irrelevant sequence were synthesized chemically and transfected instantly into Hela cells. The effects of interfering sequence on si_RALB on circRALB expression in Hela cells were detected adopting real time quantitative PCR; cell counting, MTS method, flow cytometry, cell wound scratch assay, and Transwell Chamber assay were used to study the effect of circRALB on the proliferation, the apoptosis, the translocation, and the invading capacity of Hela cells. The results showed that the efficiency of instant transfection was at (89.67±3.1)%. The expression of circRALB in si_nc group transfected with interfering sequence si_RALB was at (47.3±1.5)%, with significant statistic difference; and the cell proliferation activity of si_RALB transfected group obviously lower than that of si_nc control group with significant statistic difference; the cell apoptosis rate of si_RALB was (9.17±1.17)%, and the cell apoptosis rate of si_nc control group was (7.2±0.32)% and the cell apoptosis of the blank control group was (5.63±0.31)%, even though the cell apoptosis rate of si_RALB cells larger than the cell apoptosis rate of si_nc control group, yet had no statistic significance; comparison of distance between the scratches of si_RALB group and si_nc group the shrinkage was not obvious; the number of cell penetration of si_RALB transfected group was (21.33±2.19) significantly lower than that of si_nc group (45.33±1.76) and that of blank control group (52±1.53). Therefore, the expression of circRALB of chemically synthesis could be significantly reduced after the transfection with interfering sequence into Hela cells. And the down regulation of circRALB expression could significantly inhibit the ability of the proliferation, migration, and invasion of cervical cancer cell line of Hela cells, but had no significant effect of the apoptosis. The results of the study suggested that the expression of cirRALB related to the biological behaviors of cervical cancer cell, and may become an effective treatment target of cervical cancer.
